SLAMseq Kinetics Kit – Catabolic Kinetics Module, 24 preps

The SLAMseq Kinetics Kit – Catabolic Kinetics Module (Cat. No. 062) is designed for measuring RNA degradation kinetics in cultured cells. The RNA in cultured cells is labeled with S4U for up to 24 h, followed by treatment of cells with unlabeled uridine. In this way, existing RNA is labeled with S4U while newly synthesized RNA is unlabeled.

The Catabolic Kinetics Module provides reagents for S4U labeling of RNA in cultured cells, additional unlabeled Uridine, and iodoacetamide for the alkylation of incorporated S4U nucleotides in total RNA. The resulting total RNA can be used for RNA-Seq library preparation – no additional biochemical isolation or pull-down is required!

For cost-efficient analysis of RNA kinetics, we recommend using QuantSeq 3’ mRNA-Seq for preparing RNA-Seq libraries from total RNA generated from SLAMseq experiments. QuantSeq provides a user-friendly workflow, enabling samples from multiple timepoints and in replicates to be multiplexed in a single sequencing lane or run. SLAMseq can also be combined with other Lexogen library prep kits, including CORALL for whole transcriptome sequencing, LUTHOR HD for high-definition single-cell RNA Sequencing, and our small RNA-Seq solution.

The kit is applicable for 24-well cell culture format but can be scaled up or down to suit different culture vessel sizes and volumes. SLAMseq is compatible for cell suspensions, adherent cells, and 3D-scaffold cell cultures.
